I am using EG 5.1 query builder for the first time, i am not able to save the transposed process.Able to select the options data-->transpose and able to select the column but after that i am not getting the save and run button activated.

Are you sure that you moved the variable you want to transpose into the transpose variable(s) cell on the upper right portion of the menu?  The save and run buttons won't activate unless there is at least one variable assigned to that cell.
