data test;
format time1 time2 diff time8.;
time1 = input('09:15',time5.);
time2 = input('17:40',time5.);
diff = time2 - time1;
output;
time1 = input('09:40',time5.);
time2 = input('16:50',time5.);
diff = time2 - time1;
output;
run;

proc print data=test noobs;
run;

Result:

  time1       time2        diff

9:15:00    17:40:00     8:25:00
9:40:00    16:50:00     7:10:00

Are the pairs in one or separate strings?

convert char time as var="09:15" using time_1 = input(var,time5.);

to calculate the difference just do: diff = intck('minute',time_1,time_2);
